Replication of Adenovirus Type 12 and Formation of its Antigen.


Abstract
Approximately 40 separate adenoviruses have been isolated from man. Adenoviruses replicate in the nuclei of infected cells. They are 70 nm to 90nm in diameter, and each capsid is composed of 252 capsomers.
All members appear to have a core composed of single molecule of DNA, and a common complement fixing structural virus antigen.
Adenoviruses infection in man produce acute pharyngitis, trachcobronchitis, bronchopneumonia or conjunctivitis.
Several members of this group, ineluding types commonly associated with respiratory disease in man, are capable of producing malignant tumors in young hamster and a few types have been chown to be oncogenic in young rat.
Previous reports involving production and release of adenoviruses and its antigen have been carried out.
The present report represents a continuation of previous study.
To obtain evidence concerning the relationghip between CF antigen and infectious virus, investigation of adenoviruses type 12 of this group was undertaken.
For practical consideration it was disirable to investigate of growth characteristics and its antigen induction in HeLa, FB and MK cells(permissive cells).
@ES Results were as follows
@EN 1. CF antigen and infectious adenoviruses type 12 viruses were produced independently and the CF antigen was not a precuser to infectious adenoviruses type 12.
2. Intracellular Cf antigen had eclipse period of 16-22hrs and intracellular infectious virus was detected 16-23hrs after infection of HeLa, FB and MK cells.
3. HeLa and MK cells infected with adenoviruses type 12 were capable of continued antigen production despite early developement of maximum cytopathogenic effect.
4. This study with adenovirus type 12 indicated this virus has similar growth characteristics in HeLa, KB and MK cells.
